چکیده

5-Fluorouracil, at lop4 M concentrations, inhibited ribosomal RNA maturation only slightly in Novikoff hepatoma cells growing in suspension culture. On a molar basis, 5-fluorouridine was a much more potent inhibitor of the maturation process. The difference between the effectiveness of 5-fluorouracil and 5-fluorouridine is probably a result of the greater transport and incorporation of the latter into RNA. The nucleoside analogue strongly inhibited the production of mature 18 S and 28 S ribosomal RNA, while apparently only slightly inhibiting the formation of the initial 45 S ribosomal precursor RNA or the 38 S ribosomal RNA intermediate. Inhibition of 32 S ribosomal precursor RNA production was greater than that of the larger precursor molecules, but less than that of the mature ribosomal RNA molecules. S-Fluorouridine did not affect the methylation of 45 S ribosomal precursor RNA, indicating that the analogue exerts its inhibitory effect on some other aspect of the ribosomal RNA maturation process.

The inhibition of ribosomal RNA synthesis and maturation in Novikoff hepatoma cells by 5-fluorouridine.

The inhibition of ribosomal RNA (rRNA) maturation by 5-fluorouridine (FUrd) in Novikoff hepatoma cells appears to depend upon the incorporation of the analog into the 45 S rRNA precursor. Precursor synthesized in the presence of FUrd is not processed into mature rRNA, but precursor synthesized in the absence of the analog is processed normally after the addition of the drug. The effect of FUrd ...

Inhibition of ribosomal RNA maturation in Novikoff hepatoma cells by toyocamycin, tubercidin, and 6-thioguanosine.

The purine nucleoside analogs, toyocamycin, tubercidin, and 6-thioguanosine, all inhibit ribosomal RNA maturation in cultured Novikoff hepatoma cells. While processing of the 45 S RNA to the 38 S RNA continues, formation of the mature 28 S RNA and the mature 18 S RNA is markedly inhibited by the addition of any of the analogs. Labeling of the 32 S RNA intermediate precursor is inhibited to a le...

Effect of 5Fluorouracil on RNA Metabolism in Novikoff Hepatoma Cell&

The resistance of a Novikoff hepatoma cell line (Ni -Si I FdUrd) to 5-fluorouracil (FUra) was reversed by the inclusion of inosine in the culture medium. As the concentration of inosine in the medium was increased, there was a marked increasein the uptakeof [14C]FUra nd conversionto nucleo tides with a corresponding increase in the incorporation into RNA. While FUra alone had no effect on this ...

Metabolism of 5-fluorouracil in sensitive and resistant Novikoff hepatoma cells.

N1-S1/FdUrd Novikoff hepatoma cells, which lack thymidine kinase activity, are resistant to 5-fluorouracil (FUra) as well as 5-fluorodeoxyuridine (FdUrd), suggesting that the pathway, FUra leads to FdUrd leads to FdUMP, is utilized for the conversion of FUra to FdUMP. However, the inhibition of thymidylate synthetase activity, the presumed target of FdUMP, by 1 X 10(-4) M FUra in intact N1-S1 N...
